SOLUTIONS

The recipes given below are for general use solutions and reagents. Specialized solutions that are used in only one protocol are included within the protocol.

Ampicillin

Add to LB or LB agar at concentration of 75µg/ml

Tetracycline

Add to LB or LB agar at concentration of 75µg/ml

Denhardt’s solution

10 g Ficoll 400
10 g polyvinylpyrrolidone
10 g BSA
H2O to 500 ml

Filter and store at -20°C in 25 ml aliquots

Grape agar plates

40 ml grape juice
160 ml water
4 g bacteriological agar

  • boil to dissolve agar
  • cool to 50°C
  • add 1 ml glacial acetic acid and 2 ml 95% ethanol

Homogenization Buffer

100mM Tris-HCl, pH 9.1
100mM NaCl
200mM sucrose
50 mm EDTA
0.5% SDS

Loading Buffer for Agarose Gels (10X)

20% (w/v) Ficoll 400
0.1
M Na2 EDTA, pH 8.0
1.0% (w/v) SDS
0.25% (w/v) bromphenol blue
0.25% (w/v) xylene cyanol 

LB

10 g tryptone
5 g yeast extract
5 g NaCl
1 ml 1M NaOH

LB agar plates

1.5% bacteriological agar in LB broth

PBS (10X)

80 g NaCl
2 g KCl
11.5 g Na2HPO4*7 H2O
2 g KH2PO4

PBS-T

1X PBS with 0.5% Triton-X 100

SOC medium

0.5% yeast extract
2% tryptone
10 mM NaCl
2.5 mM KCl
10 mM MgCl
20 mM MgSO4
20 mM glucose

50X TAE:

242 g Tris base
57.1 ml glacial acetic acid
37.2 g Na2EDTA-2H20
H2O to 1 liter
TENS Solution
